The subway is two useful lines and a short stub, and it stops a little after one; the overnight buses and streetcars take over on the main corridors until the morning. Streetcars work the east–west arteries and are slower than they look. Downtown is walkable in the ordinary sense for half the year and, in the cold half, walkable underground — the PATH connects a large part of the core without anyone going outside, which is a genuinely useful thing to know in February.
The year splits hard. Patio season runs roughly May to September and is a real social institution rather than a turn of phrase; winter is properly cold, with wind off the lake and snow that arrives in quantity rather than in flurries. Autumn is the best-looking stretch, and September brings the film festival, which fills the downtown hotels and the better tables for its ten or eleven days. February and March are the quiet months and the easiest to arrange at short notice.

Enough to build the evening around. Between December and March the outdoor half of any plan gets short, and downtown the underground walkways mean a good deal of it need not be outdoors at all.
Queen West or Leslieville for something informal, Yorkville if the evening is meant to be an occasion. The Distillery District is the obvious answer in December and correspondingly full.
For the two main axes, yes, and the overnight buses cover the hours after it closes. Anything off those lines is a streetcar or a car, and both take longer than the distance suggests.
